Kansalaisvapauksien sekä oikeus- ja sisäasioiden valiokunnan toimivaltaan kuuluvat perussopimuksissa ja Euroopan unionin perusoikeuskirjassa vahvistettujen kansalaisten oikeuksien, ihmisoikeuksien ja perusvapauksien suojelu sekä vähemmistöjen suojelu unionin alueella. Valiokunta tarkastelee myös henkilötietojen suojaamista koskevia kysymyksiä. Lisäksi sen vastuualueeseen kuuluvat turvapaikka- ja maahanmuuttosäännöt, ulkorajojen yhdennetty valvonta sekä poliisiyhteistyö ja oikeudellinen yhteistyö rikosasioissa.
